Stainless steel needle electrochemical electrode · minimally invasive sensing technology
Acupuncture and moxibustion needles have been used for thousands of years in China to stimulate specific points of the human body to treat diseases. But in recent years, this small stainless steel needle has taken on a new identity - the "working electrode" of electrochemical sensors.
Conventional stainless steel microneedle sensor components often require time-consuming laboratory manufacturing processes. However, researchers found that the commercially available stainless steel acupuncture and moxibustion needle itself is an almost ideal electrode substrate - exquisite in size (needle diameter can reach 0.15~0.5mm), high in mechanical strength, low in cost and good in conductivity. Through surface functional modification, an ordinary acupuncture and moxibustion needle can become a micro electrochemical sensor that can detect glucose, heavy metal ions, neurotransmitters and other targets.
Core breakthrough:The stainless steel needle type electrochemical electrode combines the geometric advantages of acupuncture and moxibustion needles or microneedles with the surface function of electrochemical sensing - the needle body acts as a conductive substrate and mechanical support, and the needle tip becomes a sensing surface after functional modification, so that an ordinary metal needle has the dual capabilities of "puncture entry" and "chemical detection" at the same time.
The core advantage of stainless steel needle electrodes is rooted in the comprehensive performance of stainless steel materials themselves.
The needle diameter can reach 0.15-0.5mm, which is not easy to break and can penetrate the stratum corneum, sediment surface or plant tissue of the skin. The research team once processed acupuncture and moxibustion needles with a diameter of 160 μ m into 1mm microneedles, arranged them into 5 × 8 arrays, and built wearable microneedle sensing devices.
Chromium (Cr) in stainless steel forms a dense oxide film in air, endowing the electrode with stable passivation behavior in acidic media. It exhibits stable electrochemical response in 0.25mol/L H ₂ SO ₄.
Good conductivity meets the requirements of electrochemical testing, reduces ohmic drop, and improves signal-to-noise ratio. The SS304 needle electrode exhibits extremely shallow non Faraday background current in H ₂ O ₂ detection, effectively attenuating background noise.
Complies with ISO 10993 biocompatibility standard and can be used for in vivo testing; Models such as 304, 430, 316L have low prices and are suitable for one-time use. 304 stainless steel has excellent comprehensive performance and has become the preferred material for biomedical electrodes.
The sensitivity of a bare stainless steel needle electrode is limited - it is just a conductive needle and does not have the ability to selectively recognize specific target molecules. Surface functionalization modification is a key step in transforming it into a true electrochemical sensor. The current mainstream modification strategies include three types:
Electrodeposited CS-PVP substrate, followed by self-assembly of silver nanoparticles. The detection limit for NO ∝⁻ is 1.2 μ M, and the detection limit for Cu ² ⁺ is 15nM. Provides catalytic activity and specific surface area.
Etching forms a porous structure and electrodeposits gold nanoparticles. The linear range of Cu ² ⁺ detection is 0.1-1000nM, the detection limit is 0.1nM, and the spatial resolution is 4mm.
The PDA nanoparticle layer has reducibility and can reduce chloroauric acid ions to generate Au NPs, providing binding sites. Used for highly selective detection of formaldehyde in fish.
More cutting-edge research also involves layer by layer composite of various materials. For example, a multi-layer structure of Pd, Ni, Au, and Ni was sequentially deposited on a stainless steel needle (Ni Au Ni Pd/SS needle), achieving non enzymatic glucose detection with a sensitivity of 31.51 μ M/mM and a detection limit as low as 9 μ M. The layer by layer self-assembly of gold nanoparticles with multi walled carbon nanotubes and polydopamine has also been used for simultaneous detection of dopamine, acetaminophen, and chlorpromazine. An electrochemical biosensor for non enzymatic glucose detection was realized by electrodepositing gold nanoparticles and platinum nanoparticles on the surface of stainless steel acupuncture and moxibustion needles in turn.

The application of stainless steel needle electrochemical electrodes is moving from the laboratory to the forefront of industry, covering multiple fields such as biomedical, environmental monitoring, and neural regulation.
More than 500 million people suffer from diabetes worldwide. A glucose sensor based on SS304 needle electrode showed linear correlation (R ²=1.000) within the range of 1-20mM, with a reproducibility RSD of 4.4% -6.8%. Wearable microneedle arrays have been used for continuous monitoring of interstitial fluid, achieving painless real-time detection. Stainless steel microneedles modified with GOx chitosan complex and coated with hydrogel coating can selectively detect glucose in skin simulated interstitial fluid.
Au NPs functionalized needle electrode has a detection limit of 0.1nM for Cu ²+; porous gold nano material functionalized acupuncture and moxibustion needle is used for the detection of copper in seawater (detection limit 0.24nM). In situ analysis of vertical concentration distribution of Cu ²+in sediment pore water has been realized with a spatial resolution of 4mm. The sensor based on acupuncture and moxibustion needles is also used to detect cadmium ions in aquatic products.
Used for recording and regulating neural signals. With reasonable surface modification, the 1kHz impedance can be controlled at 50 – 150 Ω, and the signal noise ratio is>85dB. The minimally invasive needle sensor based on stainless steel acupuncture and moxibustion needles has been used to monitor the oxygen level in brain tissue in real time.
The stainless steel microneedle tip is used to construct a graphene PEDOT solid-state transfer layer and an ion selective sensitive membrane, achieving rapid in-situ detection of Mg ² ⁺ in crop stems.
The diameter of conventional acupuncture and moxibustion needles is 0.15 – 0.5mm; the wearable microneedle array can be as fine as 160 μ m. Finer needles mean less trauma, but require higher mechanical strength.
304 has excellent comprehensive performance and is the preferred choice for biomedical applications; 316L has better corrosion resistance and is suitable for corrosive environments such as seawater.
Glucose detection requires enzymatic (GOx) or non enzymatic (Ni Au Ni Pd, etc.) catalytic layers; Heavy metal detection requires functional layers such as Au NPs; Neural recording requires a low impedance, high signal-to-noise ratio interface.
Insulation is required for the needle rod, exposing only the sensing surface of the needle tip. Common methods: PVC pipe wrapping, polymer coating, and sealing with silicone rubber at the joint.
The essence of stainless steel needle type electrochemical electrodes is to endow an ordinary metal needle with the ability of "perception" - it retains the mechanical advantages of the needle (minimally invasive, precise, and puncturable), and obtains the "chemical sense of smell" of electrochemical sensing through surface functionalization.
It is not a single product, but a highly customizable technology platform with selectable needle diameters, materials, modification schemes, and adjustable detection targets. From continuous blood glucose monitoring to heavy metal detection in seawater, from neural regulation to in vivo analysis of plants, stainless steel needle electrodes are bringing electrochemical detection from the laboratory to the field, from in vitro to in vivo, and from macroscopic to microscopic.
